Will Ferrell was great in Old School and Wedding Crashers. Ma...meatloaf!!! &^%$!!!
Still love Chris Farley in Tommy Boy.
Not sure if William Macy role in Fargo counts as comedic, but he there's a dark humor in his performance.

Not in movies, but Michael Richard's Cosmo Kramer still cracks me up. He's useless in any other venue, so it's definitely the character I enjoy.
 
George Clooney from "O Brother..." and Jeff Bridges from "Big Lebowski" are 2 that come to mind.
 
The scene in Slap Shot where the cops come to arrest the Hanson brothers in the locker room and Paul Newman is telling the cops to take them, he can't handle them anymore, and then closes the door and encourages the Hanson brothers and the rest of the Chiefs, is a classic and one of my favorites as a kid. Paul Newman as Reggie Dunlop was great!

Also mentioned above: Stripes-Bill Murray and Harold Ramis, Caddyshack-Chase, Dangerfield, Knight Fletch-Chevy Chase, Tommy Boy with Spade and Farley.

Reggie Hammond (might have been more of a drama, but Murphy was funny as hell....Roooooooooxaannnnne!). Eddie Murphy had a great run with 48 Hours, Trading Places and Beverly Hills Cop, loved his characters in all of those movies. He also had decent characters in Coming To America and The Golden Child.

Not sure why this just popped in my head but for anyone with teenage kids, the Madagascar movies were probably in your rotation at some point. Those penguins cracked me up every time they were on screen.

I only listed a few movies that I remember watching when I was between 10-14 years old, because I think they are shown on TV still to this day. There a bunch of others out there and I could probably start with characters from Pulp Fiction and Reservoir Dogs, and I can't mention comedic characters without mentioning Blazing Saddles.

This is kind of a layup answer for sure, but all the characters from Anchorman are pretty awesome.

Bill Murray in Ground Hog Day is great.

Michael Cera in This is the End was funny

I like Chevy Chase in Funny Farm.

I could go one forever with this list...and I'm more than sure if I thought about it longer I could come up with some much better ones. I'm trying to think of some under the radar ones though.

Bill Murray in What About Bob cracks me up.

Not a movie, but my all time favorite comedic character is Kenny Powers. I must have cried from laughing so hard in a majority of the episodes
